Abstract

Penelitian ini bertujuan untuk mengkaji pengaruh ekstrak pasak bumi terhadap proporsi jantanisasi ikan guppy dan menentukan kosentrasi ekstrak batang pasak bumi yang terbaik untuk menghasilkan ikan guppy jantan. Metode dilakukan menggunakan larva ikan guppy yang baru menetas berumur 5 hari. Pembuatan ekstrak pasak bumi dengan methode maserasi, pembuatan ekstrak dilakukan dengan merendam 500 gram serbuk ke dalam etanol (96%) sebanyak 3.2 liter diaduk dengan stirer selama 3 jam kemudian didiamkan selama 24 jam. Filtrat diuapkan dengan rotary evaporator dengan suhu 60°C sampai diperoleh ekstrak kental. Proses ekstraksi dilakukan kurang lebih sebanyak 1 kali. Metode penelitian ini menggunakan Rancangan Acak Lengkap (RAL) 5 perlakuan 3 ulangan. Perendaman pasak bumi diberikan selama 24 jam dengan dosis berbeda, pemeliharaan setelah perendaman selama ±60 hari. Hasil dari penelitian menunjukkan bahwa persentase ikan guppy jantan perlakuan berkisar antara 73,02%-96.80%, persentase ikan guppy betina berkisar antara 9,58%-24,84%, dan persentase ikan guppy interseks berkisar antara 0,00%-4,41%. Kosentrasi terbaik ekstrak batang pasak bumi adalah 80 mg/L dengan nilai tertinggi 96.80% pada perlakuan E. This study aimed to examine the effect of pasak bumi extract on the proportion of male guppy fish and determine the best concentration of pasak bumi extract to produce male guppy fish. The method was carried out using newly hatched guppy fish larvae aged 5 days. The manufacture of pasak bumi extract using the maceration method, the extract was made by soaking 500 grams of powder into 3.2 liters of ethanol (96%) stirred with a stirrer for 3 hours then allowed to stand for 24 hours. The filtrate was evaporated using a rotary evaporator at a temperature of 60°C until a thick extract was obtained. The extraction process is carried out approximately 1 time. This research method uses a completely randomized design (CRD) 5 treatments with 3 replications. Soaking the pasak bumi was given for 24 hours with different doses, maintenance after immersion for ±60 days. The results of the study showed that the percentage of male guppies treated ranged from 73.02%-96.80%, the percentage of female guppies ranged from 9.58%-24.84%, and the percentage of intersex guppies ranged from 0.00%-4, 41%. The best concentration of pasak bumi extract was 80 mg/L with the highest value of 96.80% in treatment E.
